Increased immunohistochemical expression of YKL-40 in the spleen of patients with portal hypertension BJMBR
Wang,Dong; Lu,Jian-Guo; Wang,Qing; Du,Xi-Lin; Dong,Rui; Wang,Peng; Zhao,Lei; Jiang,Xue; Yuan,Li-Juan.
YKL-40 has been identified as a growth factor in connective tissue cells and also a migration factor in vascular smooth muscle cells. To a large extent, the increase of serum YKL-40 is attributed to liver fibrosis and asthma. However, the relationship of the expression and clinical/prognostic significance of YKL-40 to the splenomegaly of patients with portal hypertension is unclear. In the present study, the expression of YKL-40 was studied by immunohistochemistry in 48 splenomegaly tissue samples from patients with portal hypertension and in 14 normal spleen specimens. All specimens were quickly stored at -80°C after resection. Primary antibodies YKL-40 (1:150 dilution, rabbit polyclonal IgG) and MMP-9 (1:200 dilution, rabbit monoclonal IgG) and...

Inmunohistochemical Evaluation of Estrogen Receptors Alpha and Beta in Normal Inferior Turbinate Mucosa International Journal of Morphology
Millas,Ieda; Liquidato,Bianca Maria; Dolci,José Eduardo Lutaif; Macéa,José Rafael; Fregnani,José Humberto Tavares Guerreiro; Meceles,Lenira Rocha.
It has been postulated that the nasal mucosa, like other human tissues, is affected by a complex interactive network of neuropeptides, cytokines, allergic and inflammatory mediators and hormones such as estrogen, in which associations between symptoms (e.g. nasal stuffiness and coryza) and hormonal variations deriving from pregnancy, use of hormonal contraceptives and menstrual cycle phases are observed. The objective is evaluating the presence of specific estrogen receptors (types alpha and beta) in inferior turbinate mucosa in healthy subjects without nasal symptoms. Samples of nasal inferior turbinate were removed from patients undergoing aesthetic nasal surgery, and analyzed using hematoxylin-eosin staining, followed by immunohistochemical preparations...

Insertion of the LINE-1 element in the C-MYC gene and immunoreactivity of C-MYC, p53, p21 and p27 proteins in different morphological patterns of the canine TVT Arq. Bras. Med. Vet. Zootec.
Lima,C.R.O.; Faleiro,M.B.R.; Rabelo,R.E.; Vulcani,V.A.S.; Rubini,M.R.; Torres,F.A.G.; Moura,V.M.B.D..
ABSTRACT The canine transmissible venereal tumor (TVT) affects the external genitalia of dogs by the natural transplant of viable tumor cells. Thus, this research aimed to diagnose and characterize TVT morphological patterns, identify the insertion of the LINE-1 element in C-MYC gene, by means of the polymerase chain reaction (PCR), and evaluate the immunohistochemical expression of C-MYC, p53, p21 and p27 proteins. The relationship between C-MYC and p53 proteins and their interference on the expression of p21 and p27 were also studied. For that, 20 samples of naturally occurring TVT were used, subjected to cytopathological, histopathological and immunohistochemical analysis, and to molecular diagnosis of neoplasia. The increased tissue expression and the...

Firmino,M.O.; Frade,M.T.S.; Alves,R.C.; Maia,L.Â.; Olinda,R.G.; Ximenes,R.G.; Souza,A.P.; Dantas,A.F.M..
ABSTRACT We report a case of intestinal intussusception secondary to enteritis caused by Pythium insidiosum infection in a 1-year-old mixed breed bitch that died after a 30-day history of bloody diarrhea, anorexia, weight loss, listlessness, dehydration, and pale mucous membranes. Necropsy revealed two areas of intussusception, one jejunum-jejunal and one ileum-cecum-colic. The first intussusception showed slight congestion of the intestinal loop, which was resolved by manual traction, while the second intussusception exhibited thickening, irregular yellow spots, and transmural congestion. Histologically, the jejunum-jejunal segment had ischemic lesions secondary and ileum-cecum-colic intussusception there was also necrotizing, pyogranulomatous enteritis...

Intraoral Mucoepidermoid Carcinoma of Salivary Glands: Lack of Association Among Clinicopathological Features and Immunoexpression of c-erbB-2 in 29 Cases International Journal of Morphology
Bernardes,Vanessa Fátima; Ramos-Jorge,Maria Letícia; Vieira Carmo,Maria Auxiliadora; Cardoso,Sérgio Vitorino; Alves Mesquita,Ricardo; Ferreira Aguiar,Maria Cássia.
The association among clinicopathological features and c-erbB-2 oncoprotein expression was evaluated in twenty-nine cases of intra-oral mucoepidermoid carcinoma (MEC). MEC was prevalent in the female gender (79.3%), tumors were more frequent in ages between 21 and 40 years (48.3%), and the palate was the most commonly affected site (72.4%). Microscopically, 27 cases (93.1%) were classified as low grade of malignancy. The c-erbB-2 expression was considered positive in 9 (31%) cases and no significant association (p>0.05) was found among protein expression and gender ñor between patient age and site or histological grade of the lesión. c-erbB-2 expression in MEC may reflect intrisinc biologic properties of salivary gland neoplasms and may be linked to...

Localization of Cannabinoid Receptor 1 (CB1) in Submandibular and Sublingual Salivary Glands of Mice throughout Postnatal Development International Journal of Morphology
Hipkaeo,Wiphawi; Watanabe,Masahiko; Kondo,Hisatake.
In view of the fact that human marijuana users often show dry mouth symptom, the present study was attempted to examine the localization of CB1, which was originally identified in brain, in the submandibular and sublingual salivary glands of postnatal developing male mice by immunohistochemistry. In submandibular gland, CB1-immunoreactivity was positive in a majority of acinar cells in forms of granular appearance in their apical cytoplasm, while it was negative in the ducts at newborn stage. The immunoreactivity decreased in the acinar cells at P1W and no immunoreactivity was detected in the acinar cells at P3W and thereafter. The immunoreactivity was positive in ductal cells at P3W and it remained positive thereafter until P8W stage. The immunoreaction...

Lymphatic fluctuation in the parenchymal remodeling stage of acute interstitial pneumonia, organizing pneumonia, nonspecific interstitial pneumonia and idiopathic pulmonary fibrosis BJMBR
Parra,E.R.; Araujo,C.A.L.; Lombardi,J.G.; Ab’Saber,A.M.; Carvalho,C.R.R.; Kairalla,R.A.; Capelozzi,V.L..
Because the superficial lymphatics in the lungs are distributed in the subpleural, interlobular and peribroncovascular interstitium, lymphatic impairment may occur in the lungs of patients with idiopathic interstitial pneumonias (IIPs) and increase their severity. We investigated the distribution of lymphatics in different remodeling stages of IIPs by immunohistochemistry using the D2-40 antibody. Pulmonary tissue was obtained from 69 patients with acute interstitial pneumonia/diffuse alveolar damage (AIP/DAD, N = 24), cryptogenic organizing pneumonia/organizing pneumonia (COP/OP, N = 6), nonspecific interstitial pneumonia (NSIP/NSIP, N = 20), and idiopathic pulmonary fibrosis/usual interstitial pneumonia (IPF/UIP, N = 19). Measurement of Ki-67 antigen in 159 pituitary adenomas using the MIB-1 monoclonal antibody BJMBR
Pizarro,C.B.; Oliveira,M.C.; Coutinho,L.B.; Ferreira,N.P..
Pituitary adenomas sometimes show rapid growth and recurrence, and about one third invade the structures surrounding the sella turcica. In an attempt to determine aggressive behavior at an early stage, we used the MIB-1 antibody to identify the Ki-67 antigen. The present study was designed to evaluate pituitary adenomatous tissue in terms of secretion and proliferation and to correlate the Ki-67 index with hormone phenotype and invasive behavior. Material from 159 patients submitted to one or more resections of pituitary adenomas was evaluated. Forty-two non-secretory adenomas and 43 adenomas immunoreactive for growth hormone, 19 for prolactin, 18 for growth hormone and prolactin, 16 for adrenocorticotropic hormone (ACTH), and 21 cases of...

Metastatic well differentiated squamous cell carcinoma in the prepuce of a dog: a report of clinicopathological, immunophenotypic and therapeutic approach Arq. Bras. Med. Vet. Zootec.
Auler,P.A.; Gamba,C.O.; Horta,R.S.; Lavalle,G.E.; Cassali,G.D..
This report describes a case of a well differentiated squamous cell carcinoma (SCC) of the foreskin of a dog, with metastasis in the regional lymph node. A six-year-old male intact Pit Bull dog presented a preputial ulcerated lesion with an evolution time of one year and enlarged left inguinal lymph node. Surgical resection of the preputial lesion and inguinal lymph nodes was made. The diagnosis of a well differentiated SCC was made following histopathological analysis and immunohistochemistry technique was used to confirm lymph node metastasis and to determine the histological features of the tumor. Here we demonstrated that immunohistochemistry can be an important complementary diagnostic tool to assess the prognostic features of SCC and to determine the...

MMP-1/PAR-1 signal transduction axis and its prognostic impact in esophageal squamous cell carcinoma BJMBR
Peng,Hong-hua; Zhang,Xi; Cao,Pei-guo.
The matrix metalloprotease-1 (MMP-1)/protease-activated receptor-1 (PAR-1) signal transduction axis plays an important role in tumorigenesis. To explore the expression and prognostic value of MMP-1 and PAR-1 in esophageal squamous cell carcinoma (ESCC), we evaluated the expression of two proteins in resected specimens from 85 patients with ESCC by immunohistochemistry. Sixty-two (72.9%) and 58 (68.2%) tumors were MMP-1- and PAR-1-positive, respectively, while no significant staining was observed in normal esophageal squamous epithelium. MMP-1 and PAR-1 overexpression was significantly associated with tumor node metastasis (TNM) stage and regional lymph node involvement. Patients with MMP-1- and PAR-1-positive tumors, respectively, had poorer disease-free...

Monoamines in the pedal plexus of the land snail Megalobulimus oblongus (Gastropoda, Pulmonata) BJMBR
Faccioni-Heuser,M.C.; Zancan,D.M.; Achaval,M..
In molluscs, the number of peripheral neurons far exceeds those found in the central nervous system. Although previous studies on the morphology of the peripheral nervous system exist, details of its organization remain unknown. Moreover, the foot of the terrestrial species has been studied less than that of the aquatic species. As this knowledge is essential for our experimental model, the pulmonate gastropod Megalobulimus oblongus, the aim of the present study was to investigate monoamines in the pedal plexus of this snail using two procedures: glyoxylic acid histofluorescence to identify monoaminergic structures, and the unlabeled antibody peroxidase anti-peroxidase method using antiserum to detect the serotonergic component of the plexus. Adult land...

Myelopathy associated with cervical epidural spinal metastasis of follicular-compact thyroid carcinoma in two dogs Ciência Rural
Tochetto,Camila; Rosa,Fábio Brum; Santos,Rosmarini Passos dos; Mazzanti,Alexandre; Fighera,Rafael Almeida; Kommers,Glaucia Denise.
This case report describes the clinical, histopathological and immunohistochemical findings in two dogs with myelopathy associated with metastasis of follicular-compact thyroid carcinoma. Microscopically, both primary neoplasms were characterized by polygonal cells arranged in follicles (occasionally filled with eosinophilic colloid-like material) or in sheets. The neoplastic cells had a moderately eosinophilic cytoplasm and there was moderate anisokaryosis and anisocytosis. Additionally, the neoplastic cells were positive for thyroglobulin on immunohistochemistry, confirming the follicular origin of both tumors. This is a very uncommon presentation of this pathological condition.

Neonatal handling and the expression of immunoreactivity to tyrosine hydroxylase in the hypothalamus of adult male rats BJMBR
Hermel,E.E.S.; Severino,G.S.; Cecconello,A.L.; Pereira,F.M.; Sanvitto,G.L.; Lucion,A.B..
Neonatal handling has long-lasting effects on behavior and stress reactivity. The purpose of the present study was to investigate the effect of neonatal handling on the number of dopaminergic neurons in the hypothalamic nuclei of adult male rats as part of a series of studies that could explain the long-lasting effects of neonatal stimulation. Two groups of Wistar rats were studied: nonhandled (pups were left undisturbed, control) and handled (pups were handled for 1 min once a day during the first 10 days of life). At 75-80 days, the males were anesthetized and the brains were processed for immunohistochemistry. An anti-tyrosine hydroxylase antibody and the avidin-biotin-peroxidase method were used. Tyrosine hydroxylase-immunoreactive (TH-IR) neurons were...

p53 as a prognostic factor in adrenocortical tumors of adults and children BJMBR
Sredni,S.T.; Zerbini,M.C.N.; Latorre,M.R.; Alves,V.A.F..
Mutations of the tumor suppressor gene p53 have been considered to be important determinants in several kinds of human cancer. Accumulation of p53 protein has been reported to correlate with more aggressive clinical behavior in some neoplasms. The role of p53 expression in adrenal cortical tumors (ACT) has not been elucidated but some studies have suggested its correlation with malignant behavior. Our objective was to determine if there is a correlation between the expression of immunoreactive p53 and the biological behavior of ACT. Fifty-seven ACT (21 from children and 36 from adults) were evaluated for p53 expression by immunohistochemistry in formalin-fixed paraffin-embedded tissue and analyzed in terms of outcome. The p53 parameter was utilized...

Sá,Cláudia T.; Fonseca,Linaena M. S.; Cardoso,Sérgio V.; Aguiar,Maria Cássia F.; Carmo,Maria Auxiliadora V. do.
p53 alterations have been implicated in oral carcinogenesis process. Squamous cell carcinoma is the most common malignancy of the oral cavity, the tongue being the most frequent affected site. The aim of this study was to compare p53 immunoexpression in 43 samples of oral SCC from 5 different anatomical sites: tongue, floor of the mouth, alveolar ridge/gingiva, retromolar region and other regions of the oral cavity. Thirty-three lesions (76,8%) showed positivity for p53 (mean index of 48,37%). No statistically significant differences were found between p53 expression and the different oral sites analyzed. Similarly, no differences were found when comparing tongue lesions with the remaining sites. These results suggest that it might not exist any difference...
